چکیده

Abstract The application of transcriptome analyses in forensic genetics has experienced tremendous growth and development the past decade. earliest studies main applications were body fluid tissue identification, using targeted RNA transcripts a reverse transcription endpoint PCR method. A number markers have been identified for forensically most relevant fluids tissues method successfully used casework. introduction Massively Parallel Sequencing (MPS) opened up new perspectives opportunities to advance field. Contrary genomic DNA where two copies an autosomal segment are present cell, abundant species expressed high copy numbers. Even whole sequencing (RNA-Seq) postmortem material was shown be possible. This review gives overview on applications. methods cover as well MPS approaches. High resolution being applied fluid/ determination age stains donor, estimation post-mortem interval post mortem death investigations.

Massively Parallel Sequencing and Analysis of the Necator americanus Transcriptome

BACKGROUND The blood-feeding hookworm Necator americanus infects hundreds of millions of people worldwide. In order to elucidate fundamental molecular biological aspects of this hookworm, the transcriptome of the adult stage of Necator americanus was explored using next-generation sequencing and bioinformatic analyses. METHODOLOGY/PRINCIPAL FINDINGS A total of 19,997 contigs were assembled fr...

The human salivary RNA transcriptome revealed by massively parallel sequencing.

BACKGROUND Evaluation of the salivary transcriptome is an emerging diagnostic technology with discriminatory power for disease detection. This study explored massively parallel sequencing for providing nucleotide-level sequence information for each RNA in saliva. METHODS Transcriptome profiling with the SOLiD™ system was applied to RNA isolated from unstimulated cell-free saliva (CFS) and who...

A technique for setting analytical thresholds in massively parallel sequencing-based forensic DNA analysis

Amplicon (targeted) sequencing by massively parallel sequencing (PCR-MPS) is a potential method for use in forensic DNA analyses. In this application, PCR-MPS may supplement or replace other instrumental analysis methods such as capillary electrophoresis and Sanger sequencing for STR and mitochondrial DNA typing, respectively. PCR-MPS also may enable the expansion of forensic DNA analysis metho...

Profiling the HeLa S3 transcriptome using randomly primed cDNA and massively parallel short-read sequencing.

Sequence-based methods for transcriptome characterization have typically relied on generation of either serial analysis of gene expression tags or expressed sequence tags. Although such approaches have the potential to enumerate transcripts by counting sequence tags derived from them, they typically do not robustly survey the majority of transcripts along their entire length. Here we show that ...

Development of personalized tumor biomarkers using massively parallel sequencing.

Clinical management of human cancer is dependent on the accurate monitoring of residual and recurrent tumors. The evaluation of patient-specific translocations in leukemias and lymphomas has revolutionized diagnostics for these diseases. We have developed a method, called personalized analysis of rearranged ends (PARE), which can identify translocations in solid tumors. Analysis of four colorec...
